Schrodinger developed SGR-1505, an investigational mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) inhibitor, using AI-accelerated drug discovery. MALT1 is a key mediator of the NF-κB signaling pathway and a primary driver of certain B-cell lymphomas. Schrodinger leveraged its physics-based computational platform to rapidly identify novel MALT1 allosteric inhibitors with high potency, high specificity, and favorable drug-like properties. Multiple tumor models confirmed the potent anti-tumor activity of SGR-1505 as a monotherapy and in combination with standard of care agents for B-cell malignancies. SGR-1505 is currently in Phase 1 clinical development for relapsed or refractory B-cell lymphomas including chronic lymphocytic leukemia (CLL) and Waldenstrom macroglobulinemia (WM), having received Fast Track designation for WM. Schrodinger presented initial Phase 1 clinical data at the European Hematology Association Annual Congress, showing a favorable safety profile with encouraging preliminary efficacy across a range of B-cell malignancies. The program also received orphan drug designation for mantle cell lymphoma.
